Nearly three-quarters of mutations in genes that code for proteins ? the workhorses of the cell ? occurred within the past 5,000 to 10,000 years, fairly recently in evolutionary terms, said a national consortium of genomic and genetic experts, including those at Baylor College of Medicine.

"One of the most interesting points is that Europeans have more new deleterious (potentially disease-causing) mutations than Africans," said Dr. Suzanne Leal, professor of molecular and human genetics at BCM and an author of the report. She is also director of the BCM Center for Statistical Genetics. "Having so many of these new variants can be partially explained by the population explosion in the European population. However, variation that occur in genes that are involved in Mendelian traits and in those that affect genes essential to the proper functioning of the cell tend to be much older." (A Mendelian trait is controlled by a single gene. Mutations in that gene can have devastating effects.)

The amount variation or mutation identified in protein-coding genes (the exome) in this study is very different from what would have been seen 5,000 years ago, said Leal and her colleagues in the report that appears online in the journal Nature. The report shows that "recent" events have a potent effect on the human genome.

Eighty-six percent of the genetic variation or mutations that are expected to be harmful arose in European-Americans in the last five thousand years, said the researchers.

The researchers used established bioinformatics techniques to calculate the age of more than a million changes in single base pairs (the A-T, C-G of the genetic code) that are part of the exome or protein-coding portion of the genomes (human genetic blueprint) of 6,515 people of both European-American and African-American decent. The research was an offshoot of the National Heart, Lung and Blood Institute Exome Sequencing Project.

"The recent dramatic increase in human population size, resulting in a deluge of rare functionally important variation, has important implications for understanding and predicting current and future patterns of human disease and evolution," wrote the authors in their report.

Grading Kentucky Football's Hire of Coach Mark Stoops

Kentucky has picked Florida State defensive coordinator Mark Stoops as its new head coach.

Kentucky was the first BCS program to make a coaching change this year, and the Wildcats are the first team to hire a replacement. Florida State defensive coordinator Mark Stoops landed his first head coaching gig, as he was hired as the replacement for Joker Phillips in Lexington.

Winning at Kentucky is no easy task. Although the Wildcats have an elite basketball program, the football team is just 101-146 since 1991. Kentucky has made five bowl games since 2006 but has never won more than eight games since a 9-3 season in 1985.

Needless to say, Kentucky is arguably one of the toughest BCS jobs in college football and ranks 13th out of 14 teams in the SEC in terms of desirability. Considering how tough it is to win big at Kentucky and the Wildcats' 2-10 record in 2012, Stoops has his work cut out for him over the next couple of seasons.

Positives

Strong defensive background
Considering Kentucky has never finished higher than 40th in total defense over the last five years, this is one area Stoops should be able to make an immediate impact. The Wildcats finished 13th in the SEC in scoring defense in 2012 and ranked 11th in yards allowed. This unit had a lot of youth stepping into key roles but also needs an infusion of talent. When Stoops arrived in Tallahassee in 2010, the Seminoles were coming off a horrendous season on defense. Florida State ranked 108th nationally in yards allowed and was giving up 30 points a game. However, the Seminoles made significant progress in Stoops? first season, finishing 43rd in total defense and 30th in points allowed. And in 2011 and 2012, Florida State ranked in the top 10 in scoring and total defense. The competition is tougher in the SEC, but Stoops should be able to bring some immediate improvement to this unit.

Coaching history
With a brother (Bob) that has a national championship at Oklahoma and another brother (Mike) who served as a head coach for eight years at Arizona, Mark Stoops has plenty of good bloodlines in his family. In addition to being surrounded by two brothers with head coaching experience, Stoops has coached at South Florida, Wyoming, Houston, Miami, Arizona and Florida State, so he has plenty of experience from various parts of the country and has paid his dues on each stop on the coaching ladder.

Recruiting ties
For Kentucky to be successful in football, it has to be able to pull some talent from Ohio. And this is a big advantage for Stoops, as he was born in Youngstown and is still well-connected within the state. Also, after spending three years at Florida State and time with Miami and South Florida, Stoops should have developed plenty of recruiting connections within the state of Florida. If Kentucky is going to be successful, it needs a relentless recruiter and someone who can spark some interest in the program. Mission accomplished.

Negatives

Head coaching experience
Whether or not it?s overrated when it comes to hiring a replacement, a lack of head coaching experience has to be a concern. In other SEC hires, James Franklin and Dan Mullen had no previous head coaching experience but have turned out to be solid hires. However, there?s also the other side of the coin, which includes Kentucky?s last hire of Phillips and Ron Zook at Florida.

What offense will he run?
Although Stoops should be able to bolster Kentucky?s defense, it remains uncertain what time of offense he will run. Although the Wildcats should recruit well under Stoops, they need to do something unique on offense to compete in the SEC. Before a season-ending leg injury sidelined quarterback Maxwell Smith, Kentucky was on the right track with its pass-first offense. Whether it?s a pass-first approach or a spread offense, the Wildcats need to try something different and?forget about a pro-style attack.

Building a coaching staff
In addition to finding the right fit as offensive coordinator, Stoops has to fill out a coaching staff for the first time in his career. If Kentucky makes the financial commitment to hiring good assistants, Stoops won?t have a problem building a solid staff of proven options. However, it?s never easy for a coach at his first job to piece together a staff, especially since he doesn?t have any ties to the SEC. It?s not a huge negative but something to watch as Stoops builds his coaching staff.

Final Analysis and Grade

Make no mistake: Kentucky isn?t an easy job. However, it seems the Wildcats made the right hire in picking up Stoops. The 45-year-old Ohio native is inheriting a roster that has some promising pieces on offense but needs a lot of work on the defensive side. Stoops is a high?energy guy, which should help spark some interest in the fanbase and on the recruiting trail for Kentucky. The biggest question for the Wildcats will be picking an offensive scheme that?s a little outside of the box and not a pro-style approach. Although that type of offense could work at Kentucky, Stoops needs pick a coordinator that can build a run or pass-first approach. Don?t expect Stoops to be a miracle worker, but he should turn the Wildcats into a consistent bowl team. ?

Recreational Sport ? Steps to Follow When Choosing Your Leisure ...

How do you know which recreational sport is right for you? Though this is a simple question, it may not have a simple answer. Before answering this question, it makes sense to know the definition of a recreational sport. In simple terms, a recreational sport is any sport activity or event that one engages in during leisure time. In other words, it is a sport activity or event performed primarily for entertainment and fun. Though in some cases there may be monetary rewards associated with passtime sport, it is not usually the main goal. Other reasons associated with participating in a leisure activities includes: fitness, health, socialization, competition and sport support among others.

There are several options of leisure activities available for all age categories ranging from indoor to outdoor. Making the decision on which choice to pursue can be a real challenge and this varies significantly from one person to the other. For former professional athletes, the decision may be easier especially if they continue participating in their particular former professional sports (i.e. recreational level). The big task of choosing the right recreational sport is however on beginners. In order to make the right choice(s) if you are a beginner, it is important to consider the following general guidelines:

1. Choose a sport that you understand and are passionate about: Leisure sport in a way becomes part of your lifestyle and therefore needs to be entertaining and enjoyable. Choosing something that you don?t like or have no knowledge about can be frustrating even if your friends adore it. It only makes sense to pursue something that you know its rules and regulations. However, this does not mean one cannot learn a new sport or game, but it easier to do so if you love the sport.

2. Consider your goals and objectives: As mentioned above, there are different reasons that drive people into recreational sports. Choosing a sport that is in line with your goals, for example losing weight will only enrich the whole recreational experience. Some one looking for a recreational sport to aid in cutting down some extra body weight may benefit from running, jogging, walking, hiking or swimming among other high calorie burning sport activities as opposed to playing cards or video games. On the other hand somebody looking for a recreational sport that favors socialization may benefit from the latter.

3. Facilities, tools and equipment required: Different recreational sports may require different tools, facilities or equipment or in some cases may require none at all. These tools, facilities or equipment may also vary depending on the needs of every person. For example if you want to play golf for recreation, you will definitely have to purchase a golf club, balls and perhaps shoes. You will also need an available and accessible golf course to play. In some cases if you have more than one favorite recreational sport, you may have to choose what is more convenient based on availability of facilities. It makes more sense to go running at a nearby track especially if you get off work late in the evenings than having to drive for several miles to get to a golf course (i.e. if you like both golf and running).

4. Availability of time: Since a recreational sport is a pastime sport, choosing the right one may be dictated by how much free time is at one?s disposal. Some leisure sports are more time consuming than others. Additionally some of these sports must only be played outside. Where as some sports such as running, jogging, walking, playing tennis, playing basketball or swimming offers both indoor and outdoor options, some of them, for example playing golf, hiking, kayaking, surfing among other don?t. The more time one has, the more the options to choose from and the vice versa.

5. Seasonality: Though this may not be a big deal to people living along the equatorial line geographically, it definitely affects people living in areas with winter and summer climatic seasons. Some recreational sports such as those requiring snow may only be available during winter and only in some parts of the world. So if skiing is the only favorite recreational sport that one can participate in, it means there will be nothing to do for most of the year i.e. during the other seasons. When deciding on the right sport for recreation, it is advisable to have more than one option especially in a situation where one is affected by seasonal changes.

6. Number of participants required: Where as most sports require participation of more than one person at one time, some can be pursued individually. If your schedule does not allow group or team participation, choosing a recreational sport that will enable you to participate alone is a good idea. Jogging, walking, swimming, biking and running are great examples.

As mentioned earlier, this is not an exhaustive list of guidelines, but it is only meant to give you an idea of how to pick the best sport that will make your recreational life a success. Remember the bottom line is fun and entertainment.

ComEd's Grid Modernization Work Created More Than 700 Jobs Through Third Quarter 2012

ComEd reports that work directly related to the Energy Infrastructure Modernization Act (EIMA) enacted last year created more than 700 jobs from January through September 2012, with more than 100 jobs created in the third quarter alone. These include jobs at ComEd as well as contractor and supplier positions involving a broad range of functions required to build a 21st century electric grid, from construction and engineering to consulting and administration.

"Creating the energy infrastructure of tomorrow is putting hundreds of people to work today in good-paying jobs," said Anne Pramaggiore, president and CEO, ComEd. "It gives us great pride to be able to contribute to the Illinois economy and improve the reliability of our electric grid, which helps promote business growth and development in our state."

With passage of EIMA, ComEd embarked on a 10-year, $2.6 billion program to modernize the power system in northern Illinois. The EIMA investments also have provided an important boost to local manufacturers, suppliers and service providers with the expertise needed to support the electrical system upgrade. Through October 2012, ComEd has awarded $80 million in contracts to companies providing services and products ranging from engineering to cable to smart switches.

"The grid modernization program has allowed us to create new jobs that otherwise wouldn't have existed," said Loretta Rosenmayer, founder and CEO of INTREN, a firm that is assisting ComEd with cable replacement work, storm hardening and pole replacement.

INTREN is one of several Illinois firms partnering with ComEd to bring grid modernization benefits to ComEd customers. Other firms include Meade Electric, S&C Electric, G&W Electric, Aldridge Electric and General Cable. Growing Chicago-based firms, such as PMI Energy Solutions, Primera, and MZI Group, also are realizing new opportunities through the grid modernization effort. Choctaw-Kaul, a Native American-owned distributor of safety products, recently opened a new distribution center near Midway airport as a result of the EIMA program.

ComEd's grid modernization program hit the ground running in January, with ComEd working at an accelerated pace that is already bringing benefits to customers throughout northern Illinois. Major EIMA accomplishments in 2012 to date that are improving reliability for customers include:

  • Installing approximately 380 distribution automation devices, or "smart switches," which have prevented 70,000 power outages this year.
  • Replacing or refurbishing more than 360 miles of underground residential cable ? double the pace of any previous year.
  • Replacing approximately 32 miles of mainline cable this year ? nearly three times the annual average ? for a total of 44 miles by year-end.
  • Repairing or replacing more than 2,300 utility poles ? more than 75 percent above average ? for a total of 2,500 by year-end.
  • Assessing 6,850 manholes and refurbishing 3,800.

While grid modernization work continues, the pace is slowing from earlier this year as a result of a ruling by the Illinois Commerce Commission (ICC) that reduced the funding available for the program. Following this ruling in May, the company sought rehearing on 13 issues, and the three largest were reconsidered in rehearing. Only one of these decisions was reversed by the ICC when it ruled on the rehearing Oct. 3. As a result, ComEd will face a reduction in funding of nearly $100 million per year in 2014 and beyond. These dollars cannot be recovered and subsequently reinvested into the system. ComEd has appealed the decision in court. In the meantime, ComEd is proceeding with about 75 percent of the core grid modernization programs as planned, while postponing the deployment of additional smart meters until 2015.

"We were able to make significant progress on our grid modernization so far this year because, under the EIMA legislation, we were assured that our investments in the system would be fully recovered," said Pramaggiore, who became CEO in February of this year. "We hope to reach a positive outcome soon so that we can deliver the promised reliability improvements, cost-savings and customer service benefits to our customers and create 2,000 jobs as required by the legislation and further boost the Illinois economy."

Diagram suggests Iran working on bomb

VIENNA (AP) ? Iranian scientists have run computer simulations for a nuclear weapon that would produce more than triple the explosive force of the World War II bomb that destroyed Hiroshima, according to a diagram obtained by The Associated Press.

The diagram was leaked by officials from a country critical of Iran's atomic program to bolster their arguments that Iran's nuclear program must be halted before it produces a weapon. The officials provided the diagram only on condition that they and their country not be named.

The International Atomic Energy Agency ? the Vienna-based U.N. nuclear watchdog ? reported last year that it had obtained diagrams indicating that Iran was calculating the "nuclear explosive yield" of potential weapons. A senior diplomat who is considered neutral on the issue confirmed that the graph obtained by the AP was indeed one of those cited by the IAEA in that report. He spoke only on condition of anonymity because he was not authorized to discuss the issue.

The IAEA report mentioning the diagrams last year did not give details of what they showed. But the diagram seen by the AP shows a bell curve ? with variables of time in micro-seconds, and power and energy both in kilotons ? the traditional measurement of the energy output, and hence the destructive power of nuclear weapons. The curve peaks at just above 50 kilotons at around 2 microseconds, reflecting the full force of the weapon being modeled.

The bomb that the United States dropped on Hiroshima in Japan during World War II, in comparison, had a force of about 15 kilotons. Modern nuclear weapons have yields hundreds of times higher than that.

The diagram has a caption in Farsi: "Changes in output and in energy released as a function of time through power pulse." The number "5'' is part of the title, suggesting it is part of a series.

David Albright, whose Institute for Science and International Security is used by the U.S. government as a go-to source on Iran's nuclear program, said the diagram looks genuine but seems to be designed more "to understand the process" than as part of a blueprint for an actual weapon in the making.

"The yield is too big," Albright said, noting that North Korea's first tests of a nuclear weapon were only a few kilotons. Because the graph appears to be only one in a series, others might show lower yields, closer to what a test explosion might produce, he said.

The senior diplomat said the diagram was part of a series of Iranian computer-generated models provided to the IAEA by the intelligences services of member nations for use in its investigations of suspicions that Iran is trying to produce a nuclear weapon. Iran denies any interest in such a weapon and has accused the United States and Israel of fabricating evidence that suggests it is trying to build a bomb.

Asked about the project, Iran's chief IAEA delegate, Ali Asghar Soltanieh, said he had not heard of it. IAEA spokeswoman Gill Tudor said the agency had no comment.

Iran has refused to halt uranium enrichment, despite offers of reactor fuel from abroad, saying it is producing nuclear fuel for civilian uses. It has refused for years to cooperate with the U.N. nuclear agency's efforts to investigate its program.

Iran's critics fear it could use the enriched uranium for military purposes. Such concerns grew this month when the IAEA said Iran is poised to double its output of higher-enriched uranium at its fortified underground facility ? a development that could put Tehran within months of being able to make the core of a nuclear warhead.

In reporting on the existence of the diagrams last year, the IAEA said it had obtained them from two member nations that it did not identify. Other diplomats have said that Israel and the United States ? the countries most concerned about Iran's nuclear program ? have supplied the bulk of intelligence being used by the IAEA in its investigation.

"The application of such studies to anything other than a nuclear explosive is unclear to the agency," the IAEA said at the time.

The models were allegedly created in 2008 and 2009 ? well after 2003, the year that the United States said Tehran had suspended such work in any meaningful way. That date has been questioned by Britain, France, Germany and Israel, and the IAEA now believes that ? while Iran shut down some of its work back then ? other tests and experiments continue today.

With both the IAEA probe and international attempts to engage Iran stalled, there are fears that Israel may opt to strike at Tehran's nuclear program. The Jewish state insists it will not tolerate an Iran armed with nuclear arms.

An intelligence summary provided with the drawing linked it to other alleged nuclear weapons work ? significant because it would indicate that Iran is working not on isolated experiments, but rather on a single program aimed at mastering all aspects of nuclear arms development.

The IAEA suspects that Iran has conducted live tests of conventional explosives that could be used to detonate a nuclear weapon at Parchin, a sprawling military base southeast of Tehran. The intelligence summary provided to the AP said data gained from those tests fed the model plotted in the diagram. Iran has repeatedly turned down IAEA requests to visit the site, which the agency fears is undergoing a major cleanup meant to eliminate any traces of such experiments.

The intelligence summary named nuclear scientists Mohsen Fakhrizadeh, Majid Shahriari and Fereidoun Abbasi as key players in developing the computer diagrams, adding that Shahriari and Abbasi were also involved in the Parchin testing.

Iran has for years rebuffed IAEA attempts to question Fakhrizadeh for his suspected involvement in secret programs. Shahriari was assassinated in 2010 by what Iran says were Israeli agents. Abbasi, now the head of Iran's nuclear agency, was wounded in a separate assassination attempt the same day that Shahriari was killed.

The senior diplomat, who is familiar with the Iran probe, said the agency has not yet determined any connection between Parchin and the computer models. But Olli Heinonen, who headed the IAEA's Iran investigation until 2010, said using the results of the alleged Parchin tests would "make sense as part of the design and testing of a (computer) model."

Please and Thank You : Ideas for Women Home and Family Blog


Those are the two words that even parent wants their children to use. If nothing else you want people to at least think that your children are polite. The problem is how do you get them to say Please and Thank You all the time. The first thing you must do is start early! From the moment your child is born begin using the phrases because as your child grows and learns to speak they mimic what you do.

Once your child is old enough t start handing you things and has more movement tell them Please and Thank You. For example if your child hands you his or her bottle say ?Thank You.? When they are screaming wanting something tell?them?we say ?Please.? ?Also use them?with?your spouse and other people in the home so that your child learns when it is?appropriate?to use those words.

Once your child begins to speak don?t give them an item until they say please. This will teach them how important the words are and they will naturally begin using them. That way when people come in contact with your children you will?complimented?on what a wonderful job you did of raising them.

Syrian rebels capture air base near Damascus

BEIRUT (AP) ? Syrian rebels captured a helicopter base just outside Damascus Sunday in what an activist called a "blow to the morale of the regime" near President Bashar Assad's seat of power, while the bombardment of a village near the capital killed at least eight children.

Activists said the children were killed when Syrian warplanes bombed the village of Deir al-Asafir just outside the Damascus. The village is close to suburbs of the capital that has been witnessing clashes, shelling and air raids between troops and rebels over the past months.

The Britain-based Syrian observatory for Human Rights said the bombardment of the village killed eight children. Another activist group, the Revolution Command Council said 10 children were killed when warplanes struck the village as they played outdoors.

An amateur video showed two girls lying dead in a street while the bodies of two bloodied dead boys were in the back seat of a car parked nearby. Several other wounded children were seen rushed for treatment.

Another video showed the bodies of two dead boys inside what appeared to be a makeshift hospital as others received treatment from injuries while lying on the floor.

The activist videos appeared genuine and corresponded to other AP reporting about the events depicted. Syria restricts the access of reporters.

Syria's conflict erupted in March 2011 with an uprising against Assad's regime, inspired by other Arab Spring revolts. It quickly morphed into a civil war that has since killed more than 40,000 people, according to activists. Hundreds of children have been killed since the crisis began, according to activists.

The air base takeover claim showed how rebels are advancing in the area of the capital, though they are badly outgunned, making inroads where Assad's power was once unchallenged. Rebels have also been able to fire mortar rounds into Damascus recently.

The director of the Observatory, Rami Abdul-Rahman, said rebels seized control of the Marj al-Sultan base on the outskirts of Damascus on Sunday morning. He said at least 15 rebels and eight soldiers were killed in the fighting that started a day earlier. The rebels later withdrew from the base.

Rebels appear to be trying to take over air bases and destroy aircraft in order to prevent the regime from using them in attacks against opposition forces around the country.

The rebels have no protection against the attack helicopters and fighter jets that have been blasting their positions.

Rebels have been attacking air bases in different parts of Syria, mostly in the northern regions of Idlib and Aleppo.

In the battle at the base outside Damascus, Abdul-Rahman and Damascus-based activist Maath al-Shami said rebels destroyed two helicopters with rocket propelled grenades and captured a tank. They say the base, which is on the eastern outskirts of Damascus, houses several radar positions.

"This is a blow to the morale of the regime, because it is close to the heart of the capital," said Abdul-Rahman, referring to the base that is about 15 kilometers (10 miles) from Damascus.

Al-Shami said the rebels withdrew from the base after they captured some ammunition. He said they feared counterstrikes by regime aircraft.

An amateur video posted online showed rebels walking next to two destroyed helicopters. At least three other helicopters appeared undamaged. Black smoke billowed in the distance.

Another video showed several radar posts on hills inside the large compound. Parked military trucks stood inside as rebels roamed freely.

The Observatory also reported violence in other parts of Syria, including the country's largest city of Aleppo in the north and the capital itself.

It said rebels on Sunday captured a training base for the Popular Front for the Liberation of Palestine-General Command near the Damascus suburb of Douma. The PFLP-GC is one of the Palestinian factions most loyal to Assad.

The PFLP-GC said in a statement late Saturday that the base was under attack. It said that thousands of activists and fighters who fought against Israel were trained at the base over the past 30 years.

Also Sunday, the Observatory said a bomb targeted a bus in the southern village of Othman, killing at least five people and wounding dozens. It said rebels and troops clashed in the southern region of Quneitra on the edge of the Israeli-occupied Golan Heights.

The Local Coordination Committees, another activist group, said residents found 12 bodies in the Damascus suburb of Daraya, scene of heavy clashes between rebels and government troops over the past few days.

State TV said troops clashed with al-Qaida militants in Daraya, killing some of them and confiscating a mortar that they were using in their attacks.

The station said that troops killed an al-Qaida affiliated Palestinian militant known as Abu Suhaib in the Damascus suburb of Hajira. It said his group was behind several bombings in Syria that killed and wounded dozens of people.

Assad's regime blames the revolt on a foreign conspiracy. It accuses Saudi Arabia and Qatar, along with the United States, other Western countries and Turkey, of funding, training and arming the rebels, whom it calls terrorists.

Metabolic protein launches sugar feast that nurtures brain tumors

ScienceDaily (Nov. 26, 2012) ? Researchers at The University of Texas MD Anderson Cancer Center have tracked down a cancer-promoting protein's pathway into the cell nucleus and discovered how, once there, it fires up a glucose metabolism pathway on which brain tumors thrive.

They also found a vital spot along the protein's journey that can be attacked with a type of drug not yet deployed against glioblastoma multiforme, the most common and lethal form of brain cancer. Published online by Nature Cell Biology, the paper further illuminates the importance of pyruvate kinase M2 (PKM2) in cancer development and progression.

"PKM2 is very active during infancy, when you want rapid cell growth, and eventually it turns off. Tumor cells turn PKM2 back on -- it's overexpressed in many types of cancer," said Zhimin Lu, M.D., Ph.D., the paper's senior author and an associate professor in MD Anderson's Department of Neuro-Oncology.

Lu and colleagues showed earlier this year that PKM2 in the nucleus also activates a variety of genes involved in cell division. The latest paper shows how it triggers aerobic glycolysis, processing glucose into energy, also known as the Warburg effect, upon which many types of solid tumors rely to survive and grow.

"PKM2 must get to the nucleus to activate genes involved in cell proliferation and the Warburg effect," Lu said. "If we can keep it out of the nucleus, we can block both of those cancer-promoting pathways. PKM2 could be an Achilles' heel for cancer."

By pinpointing the complicated steps necessary for PKM2 to penetrate the nucleus, Lu and colleagues found a potentially druggable target that could keep the protein locked in the cell's cytoplasm.

MEK, ERK emerge as targets

The process begins when the epidermal growth factor connects to its receptor on the cell surface.

This leads to:

* Activation of the MEK protein, which in turn activates ERK.

* ERK sticking a phosphate group to a specific spot on PKM2.

* Phosphorylation priming PKM2 for a series of steps that culminate in its binding to the protein importin, which lives up to its name by taking PKM2 through the nuclear membrane.

Once in the nucleus, the team showed that PKM2 activates two genes crucial to aerobic glycolysis and another that splices PKM RNA to make even more PKM2.

An experiment applying several kinase-inhibiting drugs to human glioblastoma cell lines showed that only a MEK/ERK inhibitor prevented EGF-induced smuggling of PKM2 into the nucleus. ERK activation then is mandatory for PKM2 to get into the nucleus.

"MEK/ERK inhibitors have not been tried yet in glioblastoma multiforme," Lu said. Phosporylated PKM2 is a potential biomarker to identify patients who are candidates for MEK/ERK inhibitors once those drugs are developed.

MEK inhibitor blocks tumor growth

The researchers also found that the two glycolysis genes activated by PKM2, called GLUT1 and LDHA, are required for glucose consumption and conversion of pyruvate to lactate, crucial factors in the Warburg Effect. Depleting PKM2 in tumor cell lines reduced glucose consumption and lactate production.

In mice, depleting PKM2 blocked the growth of brain tumors. Re-expressing the wild type protein caused tumors to grow. However, re-expression of a PKM2 mutant protein that lost its ability to get into the nucleus failed to promote tumor formation. Experiments in human glioblastoma cell lines showed the same effect.

Injecting the MEK inhibitor selumetinib into tumors inhibited tumor growth, reduced ERK phosphorylation, PKM2 expression and lactate production in mice. In 48 human tumor samples, the team found that activity of EGFR, ERK1/2 and PKM2 were strongly correlated.

Cause of PKM2 overexpression

Lu and colleagues also published a paper in Molecular Cell that revealed a mechanism for overexpression of PKM2 in glioblastoma. They found that EGF receptor activation turns on NF-KB, which leads to a series of events culminating in PKM2 gene activation.

PKM2 levels were measured in tumor samples from 55 glioblastoma patients treated with standard of care surgery, radiation and chemotherapy. The 20 with low PKM2 expression had a median survival of 34.5 months, compared to 13.6 months for the 35 patients with high levels of PKM2.

Level of PKM2 expression in 27 low-grade astrocytomas was about half of the expression found in higher grade glioblastomas.

"In these two papers, we show how PKM2 is overexpressed in tumors, how it gets into the nucleus, that nuclear entry is essential to tumor development, and identified potential drugs and a biomarker that could usefully treat people," Lu said.

Thanksgiving shopping took a noticeable bite out of Black Friday's start to the holiday season, as the latest survey found retail sales in stores fell slightly from last year.

Saturday's report from retail technology company ShopperTrak finds consumers spent $11.2 billion at stores across the U.S. That is down 1.8 percent from last year's total.

This year's Friday results appear to have been tempered by hundreds of thousands of shoppers hitting sales Thursday evening while still full of Thanksgiving dinner. Retailers including Sears, Target and Wal-Mart got their deals rolling as early as 8 p.m. on Turkey Day.

Online shopping also may have cut into the take at brick-and-mortar stores: IBM said online sales rose 17.4 percent on Thanksgiving and 20.7 percent on Black Friday, compared with 2011.

Yet ShopperTrak said retail foot traffic increased 3.5 percent, to more than 307.67 million store visits, indicating at least some shoppers were browsing but not spending freely.

"Black Friday continues to be an important day in retail," said ShopperTrak founder Bill Martin. "This year, though, more retailers than last year began their doorbuster deals on Thursday, Thanksgiving itself. So while foot traffic did increase on Friday, those Thursday deals attracted some of the spending that's usually meant for Friday."

The company estimated that shopper foot traffic rose the most in the Midwest, up 12.9 percent compared with last year. Traffic rose the least, 7.6 percent, in the Northeast, parts of which are still recovering from Superstorm Sandy.

ShopperTrak, which counts foot traffic and its own proprietary sales numbers from 25,000 retail outlets across the U.S., had forecast Black Friday sales would grow 3.8 percent this year, to $11.4 billion.

While consumer confidence has been improving, many people are still worried about the slow economic recovery, high unemployment and whether a gridlocked Congress can avert tax increases and government spending cuts ? the so-called "fiscal cliff" ? set to occur automatically in January.

And some would-be shoppers said they weren't impressed with the discounts, or that there wasn't enough inventory of the big door-busters.

"As far as deals, they weren't there," said Tammy Stempel, 48, of Gladstone, Ore. "But business have to be successful, too. I'm hoping they extend the deals through December."

She was waiting in line outside an Ikea in Portland on Saturday to buy pots and pans for her 18-year old daughter ? as a hint that it was time to move out. Stemple and her husband went shopping at two Targets, Michaels and other stores Friday, but failed to find any amazing deals, even on a flat-screen TV they wanted for themselves.

Target, Best Buy and other stores near the Ikea seemed to have few customers, and traffic at the nearby Lloyd Center Mall also was light, even for a normal weekend.

Many shoppers around the country were armed with iPads and smart phones, to check prices as well as buy.

Online auction and shoppping site eBay reported more the 2.5 times the number of mobile transactions as last year.

Online retailers worked as hard as brick-and-mortar stores to draw customers, sending each of their subscribers an average of 5.9 promotional emails during the 7 days through Black Friday. That's an all-time high, according to marketing software company Responsys.

IBM, which tracks more than 1 million transactions at 500 online retailers each day, said its data showed 24 percent of online shoppers used a mobile device to check out a retailer's site and about 16 percent of online purchases were made on a mobile device. But while total online spending rose sharply, the value of the average online order dipped about 5 percent to $181.22.

In spite of all the TV reports showing shoppers carting away laptops and giant flat-screen TVs, IBM said combined sales of consumer electronics, printers and other office supplies were up only 8 percent, with average order prices of $326.05.

Sales of appliances and other home goods rose the most, up about 28 percent from Black Friday last year. Clothing sales rose 17.5 percent, department store sales grew just under 17 percent and sales of health and beauty products rose 11 percent.

Despite the throngs in stores Thursday night and Friday, many shoppers held off until Saturday, hoping for shorter lines and less drama.

"I can't deal with all that craziness," said Miguel Garcia, a 40-year-old office coordinator who was at a Target in the Bronx, New York, on Saturday. "Compared to what I saw on TV yesterday, this is so much more comfortable and relaxed. I can actually think straight and compare prices."

Garcia was checking prices for phones and tablets at various stores, and planned to delay any purchases until Cyber Monday when he'd have a better sense of the best deals. As money has gotten tighter over the years, Garcia said he comparison shops more.

"It forces you to become a good consumer," he said.

Tanya Dunham, a 32-year-old patient coordinator representative, likewise avoided the crowds on Black Friday and was shopping at the same Target Saturday.

"I don't like to wait (in line) just to save $15 or $20," said Dunham.

For the entire holiday sales season of November and December, ShopperTrak has predicted sales should rise 3.3 percent over last year. Those two months are crucial for retailers and can account for up to 40 percent of stores' annual revenue.

112 killed in fire at Bangladesh garment factory

DHAKA, Bangladesh (AP) ? At least 112 people were killed in a fire that raced through a multi-story garment factory just outside of Bangladesh's capital, an official said Sunday.

The blaze broke out at the seven-story factory operated by Tazreen Fashions late Saturday. By Sunday morning, firefighters had recovered 100 bodies, fire department Operations Director Maj. Mohammad Mahbub told The Associated Press.

He said another 12 people who had suffered injuries after jumping from the building to escape the fire later died at hospitals. The death toll could rise as the search for victims was continuing, he said.

Local media reported that up to 124 people were killed in the fire. The cause of the blaze was not immediately clear, and authorities have ordered an investigation.

Bangladesh has some 4,000 garment factories, many without proper safety measures. The country annually earns about $20 billion from exports of garment products, mainly to the United States and Europe.

Relatives of the factory workers were frantically looking for their loved ones. Sabina Yasmine said she saw the body of her daughter-in-law, who died in the fire, but had no trace of her son, who also worked at the factory.

"Oh, Allah, where's my soul? Where's my son?" wailed Yasmine, who works at another factory in the area. "I want the factory owner to be hanged. For him, many have died, many have gone."

Mahbub said firefighters recovered 69 bodies from the second floor of the factory alone. He said most of the victims had been trapped inside the factory, located just outside of Dhaka, with no emergency exits leading outside the building.

Many workers who had taken shelter on the roof of the factory were rescued, but firefighters were unable to save those who were trapped inside, Mahbub said.

He said the fire broke out on the ground floor, which was used as a warehouse, and spread quickly to the upper floors.

"The factory had three staircases, and all of them were down through the ground floor," Mahbub said. "So the workers could not come out when the fire engulfed the building."

"Had there been at least one emergency exit through outside the factory, the casualties would have been much lower," he said.

Many of the victims were burned beyond recognition. The recovered bodies were kept in rows on the premise of a nearby school.

Army soldiers and paramilitary border guards were deployed to help police keep the situation under control as thousands of onlookers and anxious relatives of the factory workers gathered at the scene, Mahbub said. He would not say how many people were still missing.

Prime Minister Sheikh Hasina expressed shock at the loss of so many lives in the blaze and asked authorities to conduct thorough search-and-rescue operations.

The Bangladesh Garment Manufacturers and Exporters Association said it would stand by the victims' families.

Bangladesh's garment factories make clothes for brands including Wal-Mart, JC Penney, H&M, Marks & Spencer, Carrefour and Tesco.

Separately, a flyover under construction fell onto a busy market, leaving at least 14 people dead including three construction workers in southeastern city of Chittagong, an official said Sunday.

Local fire official Abdul Mannan said the concrete structure collapsed on Saturday night, and authorities recovered the bodies by Sunday morning from under the debris in the second-largest city after Dhaka.

China doesn't officially celebrate Black Friday, but Nov. 11 has become the biggest shopping day on the Chinese calendar. E-commerce sales alone reached $4.6 billion this year.?

They don?t celebrate Thanksgiving Day here, of course, but Nov. 11, has become the biggest shopping day on the Chinese calendar.

That?s because it is known as ?Singles Day? (11.11 ? geddit?) ? a type of Valentine?s Day for those without lovers, but with friends to whom they give gifts.

Online stores have cashed in on the idea, offering mouthwatering discounts on everything from cars to clothes to tempt consumers online on Singles Day, turning it into a retail bonanza? and it works.?

?I hadn?t planned to buy anything but I heard there would be great discounts so I went online to have a look,? says Liu Na, a 20-something book editor. ?I bought a blouse and a bag for myself, at 50 percent off.?

This year, e-commerce sales on Nov. 11 reached $4.6 billion, according to an estimate by the Economic Herald, a specialist daily in Shandong province.

That may not look like much compared with ?Black Friday? spending in the US (which topped $11.4 billion last year), until you take a couple of things into account:?

First, average Chinese salaries are 10 times smaller than average American salaries.

Second, this is just online. A comparable day in America might be ?Cyber Monday,? coming up after the weekend. Last year online US shoppers shelled out $1.25 billion ? little more than a quarter of what their Chinese counterparts spent two weeks ago.

The Chinese government may not be keen on the political corners of the Internet, but it loves the commercial aspect; its current five year plan for the economy foresees a fourfold jump in e-commerce?from 2010 levels to $2.9 trillion by 2015.

China has the world?s largest online population, at 538 million, and has more online shoppers than anywhere else too: On Nov. 11, some 213 million people ? nearly half of all Chinese Internet users ? visited one of Alibaba?s two retail platforms. Alibaba, which runs the two biggest e-commerce sites in China, reported sales of $2.94 billion on Nov.11.

Ten million consumers ? more than the population of Greece ? clicked on an Alibaba site in the first minute of Nov. 11, in the dead of night.

The massive Singles Day sales promotions are expected to boost the number of online shoppers even further. Ms. Liu for example, says she has normally shopped in bricks-and-mortar stores, but her Singles Day experience has converted her.

??Apart from the discounts, it?s a lot more convenient,? she says.
